Abstract

Background: Blood donation is essential for maintaining the availability of blood and its components. Different blood bag volumes, 450 mL and 350 mL, may cause physiological changes, including alterations in hemoglobin and hematocrit levels.
Objective: To determine changes in hemoglobin and hematocrit levels among donors using 450 mL and 350 mL blood bags at the Banyumas Regency Indonesian Red Cross Blood Donation Unit.
Methods: This descriptive-analytic study used a pre-test and post-test design involving 60 donors, consisting of 30 donors using 450 mL bags and 30 using 350 mL bags. Samples were selected through accidental sampling. Data were analyzed using the independent t-test.
Results: In the 450 mL group, mean hemoglobin decreased from 14.52 to 13.67 g/dL (p=0.000), while hematocrit decreased from 42.80% to 40.27% (p=0.000). In the 350 mL group, mean hemoglobin decreased from 13.98 to 13.86 g/dL (p=0.515), and hematocrit from 40.97% to 40.47% (p=0.395).
Conclusion: Hemoglobin and hematocrit decreased after donation in both groups, with statistically significant changes observed only among donors using 450 mL blood bags
